目的:探讨应用流式细胞术(FCM)在白血病免疫分型中的影响因素.方法:淋巴细胞分离液收集单个核层及血浆层细胞分别分析其表面抗原的表达量.结果:常规吸取单个核层细胞分析时细胞表面抗原的表达量与形态学及细胞化学染色结果明显不符,改用血浆层细胞测定即得到正确的表达.结论:幼稚单核细胞因比重较大可浮在血浆层中,所以如遇血浆层浑浊时应取血浆层细胞,作免疫分型.
Objective: To investigate the influencing factors of leukemia immunophenotyping by flow cytometry (FCM). Methods: Lymphocyte separation solution was used to collect mononuclear layer and plasma layer cells to analyze the expression of surface antigen. RESULTS: The expression level of cell surface antigen was significantly different from the result of morphological and cytochemical staining in routine aspiration of mononuclear cells. The correct expression was obtained by using plasma layer cells. Conclusion: The proportion of naive mononuclear cells can float in the plasma layer, so if the plasma layer is turbid, plasma cells should be taken for immunophenotyping.
