目的:确定白簕叶总黄酮提取工艺和含量测定方法。方法:采用微波辅助法提取白簕叶中的黄酮类化合物,通过正交实验确定最佳提取工艺;分别以芦丁和金丝桃苷为指标,采用紫外分光光度法在260 nm处测定白簕叶中的总黄酮。结果:最佳工艺条件:微波辐射50 s,固液比1∶20,微波功率480W,乙醇浓度30%,间歇提取2次,按最佳工艺条件提取总黄酮含量为50.04 mg/g;芦丁回归方程为:Y=33.21C+0.0106,相关系数r=0.9998,平均加样回收率为99%,RSD=2.43%(n=6);金丝桃苷回归方程为:Y=46.269C+0.0064,相关系数r=1.000,平均加样回收率为103%,RSD=0.434%(n=6)。结论:该提取工艺合理,紫外分光光度法易操作,重复性好,为测定白簕叶中总黄酮的一种切实可行的方法。
Objective: To determine the extraction process and content determination method of total flavonoids from Angelica dahurica leaves. Methods: Microwave-assisted extraction was used to extract flavonoids from Angelica dahurica leaves. The optimum extraction process was determined by orthogonal experiment. Rutin and hyperoside were used as indicators, and UV spectrophotometry was used to determine the content of flavonoids at 260 nm. The total flavonoids in the leaves. RESULTS: The optimum conditions were as follows: microwave irradiation 50 s, solid-liquid ratio 1:20, microwave power 480 W, ethanol concentration 30%, intermittent extraction 2 times, total flavonoid content was 50.04 mg/g; The regression equation was: Y=33.21C+0.0106, correlation coefficient r=0.9998, average sample recovery was 99%, RSD=2.43% (n=6); hyperoside regression equation was: Y=46.269C+0.0064 The correlation coefficient r=1.000. The average recovery was 103% with RSD=0.434% (n=6). Conclusion: The extraction process is reasonable, the UV spectrophotometry is easy to operate, and the repeatability is good. It is a practical method for the determination of total flavonoids in the white peony leaves.
