目的:建立杏香兔耳风药材中绿原酸和3,5-O-二咖啡酰基奎宁酸的含量测定方法。方法:采用HPLC法,使用Dis-covery C18柱(250mm×4.6mm,5μm),以乙腈-0.05%甲酸水溶液为流动相,流速1.0mL.min-1,梯度洗脱,检测波长为328nm。结果:绿原酸进样量在0.60~3.00μg范围内呈线性关系(r=0.9996,n=5),平均回收率为100.8%;3,5-O-二咖啡酰基奎宁酸进样量在1.30~6.52μg范围内呈线性关系(r=0.9997,n=5),平均回收率为99.4%。结论:本方法操作简便、可靠,重复性好,专属性强,可作为杏香兔耳风药材质量控制方法。
Objective: To establish a method for the determination of chlorogenic acid and 3,5-O-d-caffeoylquinic acid in Atractylodes macrocephala. Methods: The HPLC method was used, using Dis-covery C18 column (250mm×4.6mm, 5μm), acetonitrile-0.05% formic acid aqueous solution as mobile phase, flow rate 1.0mL.min-1, gradient elution, detection wavelength was 328nm. Results: There was a linear relationship between chlorogenic acid injections in the range of 0.60 to 3.00 μg (r=0.9996, n=5) and the average recovery was 100.8%; 3,5-O-dicarboxylquinic acid injections There was a linear relationship between 1.30 and 6.52 μg (r=0.9997, n=5) and the average recovery was 99.4%. Conclusion: The method is simple, reliable, reproducible, and highly specific. It can be used as a quality control method for apricot purpureus.
