在天然药物化学的研究中,用柱层析来分离植物中的各种成分是最常用的方法。在柱层析过程中,为了加强分离效果,往往采用粒度较小的吸附剂,但随着吸附剂粒度的减少,层析柱的阻力增大,致使洗脱剂流速太慢,因而耗时过长并伴有柱效降低的问题。市售的低压层析仪虽可解决此问题,但售价太高,不易普及。我们在实验过程中,设计并制做了一种简易加压装置。仅用自来水提供压力,普通玻璃柱,即可提高洗脱速度5倍左右,因而节省时间,提高了柱效,效果令人满意。现将这种装置介绍如下(装置见附图)。
