目的 :探讨肿瘤坏死因子 ( TNFα)对人鼻咽癌 CNE3细胞生长相关基因表达的影响。方法 ;体外培育的人鼻咽癌低分化上皮细胞株 CNE3,通过 MTT细胞毒试验确定TNFα的最佳抑制剂量 ,用免疫组织化学技术观察 TNFα作用 48h后 ,CNE3细胞中增殖相关的几种蛋白表达的变化。结果 :TNFα可下调 PCNA、Ki- 67、p2 1 ras、mtp5 3等蛋白的表达 ,增强 p2 1 WAF1蛋白表达。结论 :TNFα可通过影响有关细胞周期进程基因蛋白表达而抑制细胞生长
Objective: To investigate the effect of tumor necrosis factor (TNFα) on the gene expression of human nasopharyngeal carcinoma CNE3 cells. METHODS: The human nasopharyngeal carcinoma cell line CNE3 cultured in vitro was used to determine the optimal inhibitory dose of TNFα by MTT cytotoxicity assay. Immunohistochemical technique was used to observe the proliferation-related protein expression in CNE3 cells after TNFα treatment for 48h The change. Results: TNFα down-regulated the expressions of PCNA, Ki- 67, p2 1 ras and mtp53 proteins and enhanced the expression of p2 1 WAF1 protein. CONCLUSION: TNFα inhibits cell growth by affecting gene protein expression related to cell cycle progression
