目的:以益肾活血清利为法组方通过中药含药血清观察中药对HK-2细胞(人肾近端小管上皮细胞)TEMT(肾小管上皮细胞向间质肌成纤维细胞转分化)过程中的信号因子表达的影响。方法:制备中药含药血清,观察含药血清对HK-2细胞在TGF-β1诱导下EMT过程的阻抑作用。随机分为正常组、模型组、中药低剂量组、中药高剂量组、苯那普利组。观察在TGF-β1诱导下转分化过程中对Smad4及RhoA mRNA表达作用的影响。结果:中药低剂量组、苯那普利组与模型组Smad4的mRNA表达比较有显著差异;中药高剂量组RhoA的mRNA表达与模型组比较P<0.05,差异有显著性;而中药低剂量组、苯那普利组与模型组比较差异不显著。结论:在Smad4 mRNA的表达上中药低剂量组与洛汀新均显示出抑制作用,但洛汀新对RhoA mRNA的表达上无明显作用,而中药高剂量组则有明显的抑制RhoA mRNA表达的作用,说明中药的可探讨性较大,单纯通过剂量调整或许可以达到多信号途径的抑制作用而发挥拮抗EMT的过程。
OBJECTIVE: To observe the effect of traditional Chinese medicine (TCM) on TEMT (renal proximal tubule epithelial cells) TEMT (transdifferentiation of renal tubular epithelial cells to interstitial fibroblasts) In the expression of signal factors. Randomly divided into normal group, model group, low-dose Chinese medicine group, high-dose Chinese medicine group, benazapril group. To observe the effect of TGF-β1-induced Smad4 and RhoA mRNA expression during transdifferentiation. Results: Compared with the model group, the mRNA expression of Smad4 in low dose Chinese traditional medicine group and benazepril group and model group was significantly different; the expression of RhoA mRNA in high dose traditional Chinese medicine group was significantly lower than that in model group (P <0.05) , Benazepril group and model group, the difference was not significant. Conclusion: The expression of Smad4 mRNA in Chinese medicine low dose group and Lotensin all showed inhibitory effect, but Lotensin had no significant effect on RhoA mRNA expression, but Chinese medicine high dose group had significant inhibition of RhoA mRNA expression Role, indicating that Chinese medicine can be more explored, simply by adjusting the dose may be able to achieve the inhibition of multiple signal pathways and play a antagonistic EMT process.
