目的 :研究复方天佛参口服液 (TFS)逆转人红白血病耐药细胞株K56 2 /ADM耐药性的作用 ,探讨该药对K56 2 /ADM细胞膜P -糖蛋白 (P -gp)的功能及表达的影响。方法 :体外培养K56 2 /ADM细胞 ,分别用MTT法、流式细胞仪、免疫组化法等方法观察TFS对K56 2 /ADM细胞生长的抑制作用 ,检测K56 2 /ADM细胞膜上P -gp的功能及表达情况。结果 :TFS在非毒性浓度以下能逆转细胞耐药 ,与ADM合用能抑制K56 2 /ADM细胞生长 (P <0 .0 0 5 ) ;K56 2 /ADM细胞内药物浓度升高 ;K56 2 /ADM细胞上P -gp的高表达 ,经TFS作用后其表达下降 ,且 48h后完全消失 ,呈时间依赖性。结论 :TFS能逆转K56 2 /ADM耐药性 ,其机制与抑制K56 2 /ADM细胞膜上P -gp高表达有关。
