目的 观察树闙茄病镰刀菌性角膜炎房水中Th1/Th2炎性因子的变化,了解Th1/Th2炎性因子与本病炎症反应的关系。方法 茄病镰刀菌培养7d后收集真菌混悬液,调整孢子密度为10×109CFU·mL-1。清洁级树闙40只随机分为实验组30只、对照组10只,右眼为实验眼。实验组将真菌孢子混悬液50μL注入角膜基质中央,对照组注入生理盐水50μL。造模后第3天、第7天、第14天,流式细胞仪分析房水中白细胞介素(interleukin,IL)1β、IL6、IL4、IL10的水平;病理学检查观察浸润细胞类型。结果 Th1型细胞因子IL1β和IL6浓度均在造模后第7天达到高峰,造模后各时间点与对照组相比差异均具有统计学意义(均为P<0.05);Th2细胞因子IL10浓度在造模后第14天达到高峰,造模后第7天、第14天与对照组相比差异均具有统计学意义(均为P<0.05);IL4浓度仅在造模后第7天与对照组相比差异有统计学意义(P<0.05)。病理学检查结果示:炎症细胞浸润数量造模后第7天达到高峰,以中性粒细胞为主;实验组各时间点均可见菌丝平行于基质纤维生长。结论 促炎因子IL1β和IL6及抑炎因子IL10在树闙茄病镰刀菌性角膜炎炎症反应机制中发挥重要作用。
Objective To observe the changes of Th1 / Th2 inflammatory cytokines in aqueous humor of Fusarium solani konjac in tree tree so as to understand the relationship between the Th1 / Th2 inflammatory cytokines and the inflammatory response of this disease. Methods After Fusarium solani cultivation for 7 days, the fungal suspension was collected and the spore density was adjusted to 10 × 109CFU · mL-1. Clean grade tree 闙 40 were randomly divided into experimental group 30, control group 10, the right eye for the experimental eye. In the experimental group, 50 μL of fungal spore suspension was injected into the corneal stroma, and the control group was injected with 50 μL of normal saline. The levels of interleukin (IL) 1β, IL6, IL4 and IL10 in aqueous humor were analyzed by flow cytometry on the 3rd, 7th and 14th days after the model was established. Pathological examination Observe the type of infiltrating cells. Results The concentrations of IL-1β and IL6 of Th1 type cytokines all reached the peak on the 7th day after modeling. There was significant difference between the control group and the Th1-type cytokines (all P <0.05) The concentration of IL-10 cytokine reached the peak on the 14th day after modeling, and the difference was statistically significant (all P <0.05) on the 7th and 14th days after modeling compared with the control group Compared with the control group on the 7th day after modeling, the difference was statistically significant (P <0.05). The results of pathological examination showed that the number of infiltrating inflammatory cells peaked on the 7th day after neutrophil infiltration, with neutrophils as the dominant factor. The mycelium was parallel to the growth of the stromal fibers at all time points in the experimental group. Conclusion proinflammatory cytokines IL 1β and IL 6 and anti-inflammatory cytokines IL 10 play an important role in the mechanism of Fusarium keratitis inflammation.
