[Objective] To study the relevance rules of flavonoid content of domestic high-quality Ophiopogon japonicus (Thunb) Ker-Gawl. tubers at different developmental stages and determine their quality characteristics, so as to provide a scientific basis for GAP quality control and utilization of O. japonicus. [Method] O.japonicus tubers from Hangzhou City (3-year-old) and Sichuan Province (1-year-old) were graded in accordance with the fresh tuber grading standards. After enzyme killing in microwave and drying at -50 ℃, the flavonoid content of O.japonicus tubers was determined by ultraviolet spectrophotometry. SPSS17.0 version statistical software was adopted to analyze the law of correlation between the average biomass and the flavonoids content of tubers at different developmental stages. [Result] The average flavonoid content of Hangzhou and Sichuan O. japonicus tubers at different developmental stages was 0.121%-0.192% and 0.135%-0.208%, respectively. With the increase of developmental stage, the accumulation of photosynthetic products was negatively related to the flavonoids content of O. japonicus tubers (P<0.001). The regression equation was written as y1=-0.047x+0.2 (r2=0.997) for Hangzhou O. japonicus and y2=-0.050x+0.217 (r2=0.992) for Sichuan O. japonicus. [Conclusion] The accumulation of photosynthetic products is closely related to the content of secondary metabolites flavonoids in O. japonicus tubers at different developmental stages. It is scientific to divide the domestic O. japonicus tubers by the traditional grade specifications.
