以凋亡诱导剂喜树碱影响下的HL-60细胞为检测对象,对以流式细胞仪为基础建立的3种细胞凋亡定量分析方法,进行了敏感性和特异性比较,结果:3种方法对凋亡均有定性和定量作用,均能同时提供细胞周期方面的信息。加入喜树碱(CAM)120min后,低分子量DNA抽提后荧光染料(PI)染色法(Sub-G1法)和TdT酶探测DNA断端方法(TdT法)均可检测到凋亡细胞;而DNA变性后Y啶橙染色法(AO法)在加入CAM100min后,就能够证实细胞凋亡的存在。TdT法所显示的检测结果最清晰、直观。Sub-G1法操作最简便、迅速,费用最低。结论:AO法敏感性好;TdT法更适合于细胞凋亡的细胞周期特异性分析;Sub-G1法最具实用性。
To detect HL-60 cells under the influence of apoptosis inducer camptothecin, we compared the sensitivity and specificity of three kinds of apoptosis quantitative analysis methods established on the basis of flow cytometry. Results: 3 Methods of apoptosis both qualitative and quantitative effects, both can provide the cell cycle information. After adding camptothecin (CAM) for 120min, apoptotic cells could be detected by PI staining (Sub-G1 method) and TdT DNA sequencing (TdT method) after low molecular weight DNA extraction. DNA denaturation Y-orange staining (AO method) After adding CAM100min, we can confirm the existence of apoptosis. TdT method shows the most clear and intuitive test results. Sub-G1 method is the easiest, fastest and lowest cost. Conclusion: AO method is sensitive; TdT method is more suitable for cell cycle-specific analysis of apoptosis; Sub-G1 method is the most practical.
