目的探讨阴茎海绵体内注射胰岛素样生长因子1(hIGF-1)基因提高老年性大鼠阴茎勃起功能的可能性。方法9月龄>500gSD大鼠35只随机分为3组,注射PCDB-hIGF-1组15只,注射PCDB组10只,空白对照组10只。注射后1~2d测量大鼠海绵体内压(ICP),收集大鼠阴茎海绵体组织,Trizol方法提取总RNA,RT-PCR检测hIGF-1基因mRNA的表达,免疫组化检测hIGF-1蛋白表达。结果注射PCDB-hIGF-1组大鼠ICP(149±10)高于注射PCDB空质粒组(93±11)和对照组(89±12),差异有统计学意义(P<0.05);RT-PCR检测注射PCDB-hIGF-1组检测到392bp的特异性条带,注射PCDB组和对照组未检测到特异性条带;注射PCDB-hIGF-1组免疫组化均可见hIGF-1蛋白的阳性表达。结论hIGF-1基因在大鼠阴茎海绵体中有大量表达,PCDB质粒介导的海绵体内hIGF-1基因注射可部分恢复老龄大鼠的勃起功能。
Objective To explore the possibility of injecting insulin-like growth factor 1 (hIGF-1) gene into the penis into the erectile function of senile rats. Methods Thirty-five 500-day-old SD rats (9 months old> 500g) were randomly divided into three groups: 15 in PCDB-hIGF-1 group, 10 in PCDB group and 10 in blank control group. The intracavernous pressure (ICP) was measured 1 ~ 2 days after injection. The corpus cavernosum tissues were collected and total RNA was extracted by Trizol method. The mRNA expression of hIGF-1 was detected by RT-PCR. The expression of hIGF-1 protein was detected by immunohistochemistry . Results The ICP (149 ± 10) in PCDB-hIGF-1 group was significantly higher than that in PCDB-null group (93 ± 11) and control group (89 ± 12) The specific band of 392bp was detected by PCR in PCDB-hIGF-1 group, no specific band was detected in PCDB group and control group. Immunohistochemistry of PCDB-hIGF-1 group showed the positive of hIGF-1 protein expression. Conclusion The hIGF-1 gene is highly expressed in rat corpus cavernosum, and the injection of hIGF-1 gene mediated by PCDB plasmid can partially restore the erectile function of aged rats.
