柱晶白霉素(Leucomycin简称LM)的结构如图1所示,其中组分A_1、A_3的R_2侧链为异戊酰基,来自前体亮氨酸。在柱晶白霉素的生物合成过程中,存在着菌体自身产物和产物前体的反馈抑制作用。解除这些反馈抑制,就会提高抗生素的产量。本实验采用耐自身产物和耐前体类似物的复合筛选方法,筛选北里链霉菌的高产菌株。自身产物为柱晶白霉素,耐受剂量为4000u/ml,产物前体为亮氨酸(Leucine),前体类似物为4-氮杂亮氨酸和2-噻唑丙氨酸。以紫外线
