[目的]研究人参皂苷Rg3对肺腺癌NCI-H1650细胞增殖的抑制作用及对凋亡的影响。[方法]MTT法检测人参皂苷Rg3对NCI-H1650细胞增殖的抑制作用,Annexin-V-FITC/PI法检测细胞凋亡,Western blot法检测caspase-3、Bcl-2、Bax、survivin蛋白表达的情况。[结果]不同浓度人参皂苷Rg3能有效抑制NCI-H1650细胞增殖,且呈时间剂量依赖性(P<0.01)。20μmol/L人参皂苷作用NCI-H1650细胞24、48h,细胞凋亡率明显增加,与对照组相比,差异有统计学意义(P<0.01)。20μmol/L人参皂苷作用NCI-H1650细胞48h,Bcl-2、survivin蛋白表达降低、caspase-3、Bax蛋白表达增加,与对照组相比,差异具有统计学意义(P<0.01)。[结论]人参皂苷Rg3对肺腺癌NCI-H1650细胞增殖具有明显的抑制作用,呈时间剂量依赖关系,其机制可能与降低细胞Bcl-2、survivin蛋白表达,增加caspase-3、Bax蛋白表达,促进细胞凋亡有关。
[Objective] To study the inhibitory effect of ginsenoside Rg3 on the proliferation of lung adenocarcinoma NCI-H1650 cells and its influence on apoptosis. [Methods] The inhibitory effect of ginsenoside Rg3 on the proliferation of NCI-H1650 cells was detected by MTT assay. The apoptosis of NCI-H1650 cells was detected by Annexin-V-FITC / PI assay. The protein expressions of caspase-3, Bcl-2, Bax and survivin were detected by Western blot Happening. [Result] Different concentrations of ginsenoside Rg3 could effectively inhibit the proliferation of NCI-H1650 cells in a dose-and time-dependent manner (P <0.01). Compared with the control group, the apoptosis rate of NCI-H1650 cells treated with 20μmol / L ginsenoside for 24,48h was significantly increased (P <0.01). Compared with control group, NCI-H1650 cells treated with 20μmol / L ginsenoside for 48h reduced the expression of Bcl-2 and survivin protein and increased the expression of caspase-3 and Bax protein (P <0.01). [Conclusion] Ginsenoside Rg3 has a significant inhibitory effect on the proliferation of lung adenocarcinoma NCI-H1650 cells in a dose-dependent manner. The mechanism may be related to the decrease of the expression of Bcl-2 and survivin protein, the increase of the expression of caspase-3 and Bax, Promote cell apoptosis related.
