目的 :观察电离辐射诱导小鼠 S180细胞凋亡与辐射剂量、照射方式、抑瘤作用及生存期的关系。方法 :将 S180荷瘤鼠分为对照组、不同剂量组 ,以医用直线加速器 X线单次照射和分次照射。采用光镜、电镜、DNA琼脂糖凝胶电泳法 ,以及流式细胞法观察 S180细胞凋亡的形态学和生化学特征 ,并观察不同处理组的小鼠抑瘤情况及生存期。结果 :(1)单次照射、分次照射均能诱导出典型的凋亡特征。 (2 )分次照射凋亡率高于单次照射。 (3)凋亡与照射剂量、时间呈依赖关系。(4)照射有抑瘤作用 ,生存期延长。在一定剂量范围内 ,大剂量作用高于小剂量。结论 :(1)电离辐射诱导小鼠 S180细胞凋亡与照射剂量、照射方式有密切关系。 (2 )电离辐射的不同剂量对荷瘤小鼠的抑瘤作用及生存期有不同影响。 (3)电离辐射有抗肿瘤作用 ,其机制可能是通过诱导细胞凋亡实现的。 (4)研究结果将为临床上如何进行个体化放疗提供新的实验依据
OBJECTIVE: To observe the relationship between the radiation dose, the irradiation mode, the antitumor effect and the survival time of S180 cells induced by ionizing radiation in mice. Methods: S180 tumor-bearing mice were divided into control group and different dose groups, and were irradiated by single line and single-line by X-ray of medical linear accelerator. Morphological and biochemical characteristics of apoptosis in S180 cells were observed by light microscopy, electron microscopy, DNA agarose gel electrophoresis and flow cytometry. The antitumor activity and survival of mice in different treatment groups were also observed. Results: (1) Apoptotic characteristics were induced by single irradiation and divided irradiation. (2) The apoptosis rate of irradiation was higher than that of single irradiation. (3) Apoptosis and irradiation dose, the time was dependent. (4) irradiation has anti-tumor effect, prolong survival. In a certain dose range, high-dose effect than the small dose. Conclusion: (1) The apoptosis of S180 cells induced by ionizing radiation is closely related to the irradiation dose and irradiation mode. (2) Different doses of ionizing radiation have different effects on tumor-inhibiting effect and survival of tumor-bearing mice. (3) ionizing radiation has anti-tumor effect, the mechanism may be through the induction of apoptosis. (4) The results of the study will provide new experimental basis for how to carry out personalized radiotherapy in clinic
