单硝酸异山梨酯(5—ISMN)是硝酸异山梨酯主要活性代谢产物,适用于冠心病和心绞痛的防治。由于无肝脏首过效应,口服吸收完全,半衰期(t~(1/2)=5h)较长,深受临床欢迎。 根据5—ISMN分子结构中含有硝基,采用高灵敏度的电子捕获检测器、HP—5毛细管分流进样方式,选择分配行为、分子结构、挥发性及色谱行为均与5—ISMN相似的硝酸异山梨酯(ISDN)作为内标。以样品峰面积(As)与内标峰面积(Ai)的比值(s)对浓度(C)成线性关系的原理,计算血药浓度,求药动学参数。
Isosorbide mononitrate (5-ISMN) is the main active metabolite of isosorbide mononitrate, which is suitable for the prevention and treatment of coronary heart disease and angina pectoris. Due to no first pass liver effect, oral absorption completely, half-life (t ~ (1/2) = 5h) longer, deeply welcomed by the clinic. According to the 5-ISMN molecular structure containing nitryl, high-sensitivity electron capture detector, HP-5 capillary split injection mode, select the distribution behavior, molecular structure, volatility and chromatographic behavior similar to 5-ISMN nitrate Sorbitol (ISDN) as internal standard. The linear relationship between the concentration (C) and the ratio (s) of sample peak area (As) to internal standard peak area (Ai) was calculated, and the plasma concentration and pharmacokinetic parameters were calculated.
