目的探讨雨生红球藻对L-亚硝酸左旋精氨酸甲酯(L-NAME)诱导的子痫前期模型大鼠病理组织改变和氧化应激的保护作用及机制。方法采用L-亚硝酸左旋精氨酸甲酯(L-NAME)诱导产生子痫前期孕鼠,随机分为空白组(n=20)、L-NAME建模组(n=20)和雨生红球藻组(n=20)。受孕D5,雨生红球藻组大鼠灌胃1.6g/kg bw.d雨生红球藻;D10建模组和雨生红球藻组每天皮下注射L-NAME 125 mg/kg bw·d。检测血液中MDA、SOD、NOS;HE和Masson染色分析胎盘组织病理改变;免疫组化分析NF-κB(nuclear factor kappa B)、ROCK II(Rho-associated protein kinase II)、HO-1(heme oxygenase)、caspase 3蛋白表达变化。结果 L-NAME诱导大鼠产生典型的子痫前期表现,如血压、尿蛋白增高;血液中的MDA、内皮素(endothelin,ET)增高。而经雨生红球藻处理后,大鼠血压显著降低,尿蛋白减少,血液中的MDA和内皮素下降,同时雨生红球藻明显下调了胎盘组织中的NF-κB、ROCK Ⅱ、caspase 3,增加了HO-1的表达。胎盘组织中的滋养细胞数减少,纤维蛋白沉积减少,基底膜增厚现象改善。结论雨生红球藻能有效减轻子痫前期大鼠的氧化应激损伤及病理组织改变,对子痫前期疾病有一定的预防及治疗作用。
Objective To investigate the protective effect of Haematococcus pluvialis on pathological changes and oxidative stress in L-NAME-induced preeclampsia model rats and its mechanism. Methods Pregnant preeclampsia rats were induced by L-NAME, and randomly divided into blank group (n = 20), L-NAME group (n = 20) Haematococcus group (n = 20). Pregnant D5, Haematococcus pluvialis gavage 1.6g / kg bw.d Haematococcus pluvialis; D10 modeling group and Haematococcus pluvialis group daily subcutaneous injection of L-NAME 125 mg / kg bw · d . The levels of MDA, SOD and NOS in the blood were measured. The pathological changes of placenta were analyzed by HE and Masson staining. The expressions of nuclear factor kappa B, ROCK-II (protein kinase II) and heme oxygenase ), Caspase 3 protein expression changes. Results L-NAME induced typical preeclampsia in rats, such as blood pressure and urine protein increased; blood MDA and endothelin (ET) increased. After Haematococcus pluvialis treatment, blood pressure was significantly reduced, urinary protein decreased, blood MDA and endothelin decreased, while Haematococcus pluvialis significantly decreased placental tissue NF-κB, ROCK Ⅱ, caspase 3, increased the expression of HO-1. Trophoblast cells in the placenta decreased, reduced fibrin deposition, basement membrane thickening improved. Conclusion Haematococcus pluvialis can effectively reduce the oxidative stress injury and pathological changes in preeclampsia rats, and has some preventive and therapeutic effects on preeclampsia.
