目的对湖州市肠道病毒71型(EV71)进行基因特征及重组特点分析。方法对2011-2012年湖州市EV71分离株进行全基因组测序,并与其基因型及A组肠道病毒其他血清型的原型株进行全序列比对,使用Mega 4.0软件绘制系统进化树。然后利用Simplot 3.5.1软件进行相似性分析,对基因重组加以验证。结果 P1区系统进化分析显示,2011-2012年湖州市EV71株属于C4基因型的C4a亚型。P1、P2、P3区的系统进化和相似性分析证实湖州市分离株在非结构蛋白2B编码区存在EV71 C基因型和Cox A16、14、4型间重组。结论 2011-2012年湖州市EV71分离株为C4a亚型,与2004年以来中国大陆优势株流行趋势一致。EV71 C4基因型湖州市株存在型间重组现象。
Objective To analyze the gene characteristics and recombination characteristics of enterovirus 71 (EV71) in Huzhou. Methods Whole genome sequencing of EV71 isolates in Huzhou City from 2011 to 2012 was performed, and the complete genomes of the genotypes and the other serotypes of group A enterovirus strains were sequenced. The phylogenetic tree was drawn using Mega 4.0 software. Simplot 3.5.1 software was then used for similarity analysis to verify the gene recombination. Results Phylogenetic analysis of P1 showed that EV71 in Huzhou from 2011 to 2012 belonged to the C4a subtype of C4 genotype. Phylogenetic analysis and similarity analysis of P1, P2 and P3 regions confirmed that there were EV71 C genotypes and Cox A16, 14 and 4 type recombinants in the nonstructural protein 2B coding region of Huzhou isolates. Conclusion The EV71 isolates from Huzhou in 2011-2012 are C4a subtypes, which are consistent with the prevalence trends of dominant strains in mainland China since 2004. EV71 C4 genotype Huzhou strains exist between the recombination phenomenon.
