为了探讨丁酸钠对人肝癌细胞各种酶活性的影响,采用丁酸钠处理Bel-7402人肝癌细胞,测定其γ-谷氨酰转肽酶(γ-GT)、酪氨酸-α-酮戊二酸转氨酶(TAT)、醛缩酶(ALD)的比活力和细胞增殖速度。结果显示,2.5mmol/L和5.0mmol/L的丁酸钠能明显抑制人肝癌细胞增殖,并使肝癌标志酶γ-GT活性及反映肝细胞恶变或损伤的ALD活性明显降低(P<0.01,P<0.05),而使反映肝细胞分化的TAT活性明显升高(P<0.01,P<0.05)。结果表明,丁酸钠能引起人肝癌细胞γ-GT、TAT、ALD明显改变,提示这几种酶可作为肝癌细胞分化诱导的酶学指标
To investigate the effect of sodium butyrate on the activity of various enzymes in human hepatocellular carcinoma cells, Bel-7402 human hepatocellular carcinoma cells were treated with sodium butyrate and the activities of γ-glutamyl transpeptidase (γ-GT), tyrosine- Ketoglutarate transaminase (TAT), aldolase (ALD) specific activity and cell proliferation rate. The results showed that 2.5mmol / L sodium butyrate and 5.0mmol / L sodium butyrate could significantly inhibit the proliferation of human hepatocellular carcinoma cells, and markedly decreased the activity of hepatic cancer marker γ-GT and the ALD activity reflecting hepatocellular malignant transformation or injury (P < 0.01, P <0.05), while the TAT activity reflecting hepatocyte differentiation was significantly increased (P <0.01, P <0.05). The results showed that sodium butyrate can cause significant changes of γ-GT, TAT and ALD in human hepatoma cells, suggesting that these enzymes could be used as indicators of differentiation induced by hepatoma cells
