[目的]研究弓形虫感染后对雄性小鼠黄体生成素(LH)、睾酮(T)水平的影响,进一步探讨其对雄性睾丸生殖细胞的损伤特点。[方法]选用10周龄雄性BALB/C小鼠72只,随机分为:正常对照组、弓形虫感染组(1×103、2×103、4×103、8×103)及阳性对照组(环磷酰胺)。采用腹腔注射法,建立弓形虫感染模型,放射性免疫检测小鼠血清LH、T以及睾丸局部T水平;流式细胞仪检测睾丸生精细胞的凋亡情况并免疫组化SP法测定睾丸Bax、Bcl-2表达的变化。[结果]血清LH变化不明显,血清及睾丸局部的T水平降低(P﹤0.05);睾丸细胞Bcl-2表达无明显改变,而生精细胞尤其是精母细胞的Bax表达明显增加;流式细胞仪检测正常对照组未出现明显的凋亡峰,实验组的二倍体出现明显凋亡,四倍体细胞减少。[结论]弓形虫感染导致生精细胞,尤其是精母细胞的大量凋亡可能与局部T水平的降低密切相关。
[Objective] To study the effects of Toxoplasma gondii on the levels of LH and T in male mice and to explore the characteristics of the damage to male testicular germ cells. [Methods] Seventy two male BALB / C mice (10 weeks old) were randomly divided into normal control group, Toxoplasma gondii infection group (1 × 103, 2 × 103, 4 × 103, 8 × 103) and positive control group Cyclophosphamide). Toxoplasma gondii infection model was established by intraperitoneal injection. Serum LH and T levels and testicular T level were detected by radioimmunoassay. Apoptosis of spermatogenic cells in testis was detected by flow cytometry. Bax, Bcl-2 -2 expression changes. [Result] Serum LH did not change obviously, serum T and testicular T decreased (P <0.05); Bcl-2 expression of testicular cells did not change significantly, while spermatogenic cells especially spermatocytes Bax expression increased significantly; Cytometry detected normal control group did not appear significant peak of apoptosis, the experimental group of diploid significant apoptosis, tetraploid cells decreased. [Conclusion] A large number of apoptosis of spermatogenic cells, especially spermatocytes, caused by Toxoplasma gondii infection may be closely related to the reduction of local T level.
