目的探讨埃滋蛋白(Ezrin)和细胞间黏附分子1(intercellular adhesion molecules 1,ICAM-1)在结肠癌组织中的表达及意义。方法应用免疫组织化学S-P法和Western blot方法检测32例结肠癌及癌旁正常组织中Ezrin和ICAM-1的表达,分析其与结肠癌临床病理特点的关系。结果①Ezrin和ICAM-1在结肠癌组织中高表达,正常组织低表达或不表达。Western blot检测结果:Ezrin在癌组织中表达量为(2 121.0±631.5),在正常组织中为(907.8±258.3);ICAM-1在癌组织中表达量为(2 511.5±627.2),在正常组织中为(676.2±323.6),免疫组化显示二者主要表达于细胞膜和细胞质,肿瘤间质中也可见ICAM-1表达,且癌组织中的表达明显高于正常组织。②二者在结肠癌Dukes C+D期中的表达明显高于A+B期(P<0.01),在有淋巴结转移组的表达明显高于无转移组(P<0.01)。③Ezrin和ICAM-1表达成正相关(r=0.898,P<0.01)。结论 Ezrin和ICAM-1在结肠癌组织中均有表达,其表达程度与结肠癌的分期、有无淋巴结转移有关。
Objective To investigate the expression and significance of Ezrin and intercellular adhesion molecules 1 (ICAM-1) in colon cancer. Methods The expressions of Ezrin and ICAM-1 in 32 cases of colorectal cancer and adjacent normal tissues were detected by immunohistochemical S-P method and Western blot. The relationship between Ezrin and ICAM-1 expression in colorectal cancer was analyzed. Results ①Ezrin and ICAM-1 were overexpressed in colon cancer tissues, while normal tissues were either poorly expressed or not expressed. The results of Western blot showed that the expression of Ezrin in cancer tissue was (2 121.0 ± 631.5) and in normal tissues (907.8 ± 258.3), while that in ICAM-1 was (2 511.5 ± 627.2) (676.2 ± 323.6) in the tissue. Immunohistochemistry showed that the two were mainly expressed in the cell membrane and the cytoplasm. ICAM-1 expression was also observed in the tumor stroma, and the expression in the cancer tissue was significantly higher than that in the normal tissue. ② The expression of both in colon cancer Dukes C + D stage was significantly higher than that in A + B stage (P <0.01), and was significantly higher in patients with lymph node metastasis than in those without metastasis (P <0.01). ③Ezrin and ICAM-1 expression was positively correlated (r = 0.898, P <0.01). Conclusions Both Ezrin and ICAM-1 are expressed in colon cancer tissues, and the expression of Ezrin and ICAM-1 is correlated with the stage of colon cancer and the presence or absence of lymph node metastasis.
