目的 :探讨干扰素 α2 a、bcr/ abl反义寡核苷酸对慢性粒细胞白血病骨髓体外净化效果。方法 :采用体外克隆形成培养技术 ,分别用于干扰素 α2 a、bcr/ abl反义寡核苷酸不同的组合方案 ,进行体外净化 5例慢性粒细胞白血病 (CML )骨髓。结果 :联用 IFN-α2 a(1× 10 6U / L )、bcr/ abl AS- ODN(4 0 mg/ L )方案 ,对白血病细胞克隆形成单位 (L - CFU )抑制作用有显著差异 (P<0 .0 1) ,而相同条件下 ,GM- CFU及 L - CFU的集落存活率分别为 30 .0 6 %和 8.9% (P<0 .0 1)。结论 :联用 IFN -α2 a(1× 10 6U / L )、bcr/ abl AS- ODN(4 0 mg/ L )方案 ,选择性体外净化慢性粒细胞白血病细胞的效果好 ,可用于临床净化 CML骨髓
Objective : To investigate the effect of interferon alpha 2 a and bcr/ abl antisense oligonucleotides on the in vitro purification of myeloid leukemia. METHODS: In vitro cloning technique was used for different combinations of interferon α2 a and bcr/abl antisense oligonucleotides. Five cases of chronic myelogenous leukemia (CML) bone marrow were purified in vitro. Results: The combination of IFN-α2 a (1×10 6 U / L) and bcr/abl AS-ODN (40 mg / L) regimens had significant differences in the inhibitory effect on leukemic cell colony forming units (L-CFU) (P). <0. 0 1), under the same conditions, the colony survival rates of GM-CFU and L-CFU were 30.0% and 8.9%, respectively (P < 0.01). CONCLUSION: The combined use of IFN-α2 a (1×10 6 U / L) and bcr/abl AS-ODN (40 mg/L) regimens is effective for the selective purification of chronic myelogenous leukemia cells in vitro and can be used to clinically purify CML. marrow
