目的了解胃癌细胞SGC7901及其耐药细胞SGC7901/VCR中HSP90β蛋白的表达,与化疗药物敏感性的关系及降低细胞HSP90β蛋白的表达是否影响化疗药物的敏感性。方法用免疫组化法检测胃癌细胞SGC7901及其耐药细胞SGC7901/VCR中HSP90β蛋白的表达。MTT法测定SGC7901、SGC7901/VCR细胞及其相应的HSP90β蛋白低表达细胞AH-SGC7901和AH-SGC7901/VCR对化疗药物的敏感性,流式细胞仪测定细胞内药物含量。结果SGC7901/VCR细胞HSP90β蛋白的表达高于SGC7901细胞。SGC7901/VCR细胞对ADM、VCR、MMC和CTX的IC50分别为0.398、1.25、0.199和3 981.07μg/ml。SGC7901对ADM、VCR、MMC及CTX的IC50分别为1.58×10-6、1.99×10-3、3.1×10-2及630μg/ml。SGC7901/VCR相对于SGC7901细胞对ADM、VCR、MMC和CTX的耐药指数分别为2.52×105、6.28×102、6.42和6.32。AH-SGC7901和AH-SGC7901/VCR对化疗
Objective To understand the expression of HSP90β protein in gastric cancer cell line SGC7901 and its resistant cell SGC7901/VCR, and to determine whether the expression of HSP90β protein affects the sensitivity of chemotherapeutic drugs. Methods The expression of HSP90β protein in gastric cancer cell SGC7901 and its resistant cell SGC7901/VCR was detected by immunohistochemistry. The sensitivities of SGC7901, SGC7901/VCR cells and their corresponding HSP90β low expressing cells AH-SGC7901 and AH-SGC7901/VCR to chemotherapeutic drugs were measured by MTT assay. The intracellular drug content was measured by flow cytometry. Results The expression of HSP90β protein in SGC7901/VCR cells was higher than that in SGC7901 cells. The IC50 of SGC7901/VCR cells against ADM, VCR, MMC, and CTX were 0.398, 1.25, 0.199, and 3981.07 μg/ml, respectively. The IC50 of SGC7901 against ADM, VCR, MMC, and CTX were 1.58×10-6, 1.99×10-3, 3.1×10-2, and 630 μg/ml, respectively.
