目的:研究高糖环境下滋阴补肾方药左归丸对小鼠胚胎体外发育的影响。方法:体外培养小鼠胚胎,用高浓度葡萄糖抑制胚胎发育,加左归丸含药血清干预胚胎体外发育过程,用囊胚率,2-细胞率和囊胚细胞计数来评估胚胎发育的情况。结果:高糖环境抑制胚胎发育,选择40mmol/L为高葡萄糖抑制胚胎发育的浓度,与正常组比较,囊胚率下降(P<0.05),用左归丸含药血清干预高糖抑制组,囊胚率升高(P<0.05)。从囊胚细胞计数来看,高糖环境减少囊胚细胞数(P<0.05,P<0.01),左归丸含药血清进一步减少囊胚细胞数,但能有效提高囊胚率。结论:高糖抑制胚胎发育,左归丸含药血清可以提高高糖干预下受抑制的胚胎发育率,促进胚胎发育。
Objective: To study the effect of Zuogui Bu recipe (Zuyin Bushen recipe) on the development of mouse embryos in vitro under high glucose condition. Methods: Mouse embryos were cultured in vitro, embryos were inhibited by high concentration of glucose, and the serum containing Zuogui pill was used to interfere the development of embryos in vitro. The blastocyst rate, 2-cell rate and blastocyst count were used to evaluate the embryonic development. Results: The embryo development was inhibited under high glucose conditions. The optimal concentration of glucose was 40mmol / L, and the blastocyst rate was decreased compared with the normal group (P <0.05) The blastocyst rate increased (P <0.05). In terms of blastocyst count, the number of blastocyst cells was decreased in high glucose environment (P <0.05, P <0.01). Zuogui pill containing serum could further reduce the number of blastocyst but could effectively increase the blastocyst rate. Conclusion: High glucose inhibits embryo development. Zuogui pill-containing serum can increase the rate of embryo development under high glucose intervention and promote embryo development.
