目的 :探讨MTT法体外药敏试验在神经胶质瘤患者中应用的可能性。方法 :采用MTT法对50例人神经胶质瘤新鲜标本 ,进行了临床12种常用化疗药物的体外敏感性测定。结果 :采用简单机械法分离单细胞悬液 ,以每孔5×104胞量接种、1×PPC(血浆峰浓度)作用72小时为适宜的实验条件 ,试验的可评价率为98 % ;以抑制率>50%为敏感的判断标准 ,神经胶质瘤对VM26、DDP、MMC、ACLA、和VP 16较为敏感 ,敏感率在71.43%~26.53%之间 ;不同个体对同一药物及同一药物对不同个体的敏感性具有明显的差异。结论 :MTT法具有简便、快速、客观、成功率高等特点 ,在神经胶质瘤临床个体化疗上具有使用价值。
Objective: To explore the possibility of MTT assay for drug susceptibility testing in patients with glioma. METHODS: MTT assay was used to determine the in vitro sensitivity of 12 clinically used chemotherapeutic drugs in 50 fresh specimens of human glioma. RESULTS: Single cell suspensions were separated by simple mechanical method. The optimal experimental conditions were as follows: inoculation of 5 × 104 cells per well and 1 × PPC (peak plasma concentration) for 72 hours. The testable rate was 98%; A rate of >50% is a sensitive criterion. Glioma is sensitive to VM26, DDP, MMC, ACLA, and VP16, with a sensitivity rate of 71.43% to 26.53%. Different individuals have the same drug and the same drug. There is a clear difference in individual sensitivity. Conclusion : MTT assay is simple, rapid, objective, and has high success rate. It has therapeutic value in the clinical chemotherapy of glioma patients.
