目的:选用荷瘤小鼠作为研究对象,观察大黄虫丸对肿瘤细胞的诱导凋亡作用,旨在探讨其对肿瘤效应机制,为大黄虫丸抑瘤抗癌作用提供理论依据,进而创制出稳定高效、无毒副作用的抗肿瘤药物。方法:建立小鼠克洛氏(S180)肉瘤动物模型,将荷瘤小鼠分为大黄虫丸组,以大黄虫丸方41 g/kg剂量连续灌胃10 d,同时设模型对照组、正常组和西药(环磷酰胺)对照组,取实体瘤分别涂片和制成超薄切片观察肿瘤细胞形态学变化特点。结果:光学显微镜下观察大黄虫丸组小鼠肿瘤面积及瘤细胞巢明显缩小,肿瘤细胞数目明显减少;透射电子显微镜下观察大黄虫丸组小鼠肿瘤细胞体积明显变小,细胞表面微绒毛减少甚至消失,并可见凋亡小体。结论:大黄虫丸可诱导肿瘤细胞凋亡。
OBJECTIVE: To select the tumor-bearing mice as the research object and observe the effect of rhubarb Zhechong pill on the apoptosis of tumor cells in order to explore its mechanism of tumor effect and provide the theoretical basis for the anti-tumor and anti-cancer effects of rhubarb Zhechong pill A stable and efficient, non-toxic side effects of anti-cancer drugs. Methods: The animal model of murine Osteoporosis (S180) sarcoma was established. The tumor-bearing mice were divided into two groups: rhubarb and chrysanthemum pill group, and gavage at 41 g / kg for 10 days. The model control group , Normal group and Western medicine (cyclophosphamide) control group. The solid tumor was smeared and made into ultrathin sections to observe the morphological changes of tumor cells. Results: Under the light microscope, the tumor area and nests of tumor in Rhubarb group were significantly reduced, and the number of tumor cells was significantly decreased. The size of tumor cells in Rhubarb group was significantly decreased under transmission electron microscope, Fluff to reduce or even disappear, and apoptotic bodies can be seen. Conclusion: Dahuang Zhechong pill can induce tumor cell apoptosis.
