目的建立蒲公英中主要有效成分咖啡酸、绿原酸和阿魏酸的高效毛细管电泳含量测定方法。方法电泳条件:未涂层的弹性石英毛细管柱(75μm×57 cm),电解质为20 mmol.L-1硼砂溶液(pH 9.55),分离电压18 kV,柱温25℃,压力进样5 s,检测波长325 nm。结果咖啡酸、绿原酸、阿魏酸分别在0.010 2~0.081 8 mg.mL-1,0.008 1~0.065 0 mg.mL-1,0.000 6~0.004 5 mg.mL-1内呈良好线性关系,平均回收率分别98.8%,98.9%,97.8%,RSD分别为1.12%,1.21%,1.46%。结论本法专属性强,简便快速,准确度高,重复性好,适用于检测蒲公英中咖啡酸、绿原酸和阿魏酸的含量。
Objective To establish a method for the determination of caffeic acid, chlorogenic acid and ferulic acid in dandelion by high performance capillary electrophoresis. Methods Electrophoresis conditions were: uncoated elastic quartz capillary column (75 μm × 57 cm), electrolyte 20 mmol.L-1 borax solution (pH 9.55), separation voltage 18 kV, column temperature 25 ℃, Detection wavelength of 325 nm. Results There was a good linear relationship between caffeic acid, chlorogenic acid and ferulic acid in the range of 0.010 2 ~ 0.081 8 mg.mL-1, 0.008 1 ~ 0.065 0 mg.mL-1, 0.0006 ~ 0.004 5 mg.mL-1 The average recoveries were 98.8%, 98.9% and 97.8% respectively, with RSDs of 1.12%, 1.21% and 1.46%, respectively. Conclusion This method is specific, simple, rapid, accurate and reproducible. It is suitable for the determination of caffeic acid, chlorogenic acid and ferulic acid in dandelion.
