目的研究慢性宫内缺氧(CIH)对子代兔成年期腹主动脉C反应蛋白(CRP)和单核细胞趋化因子1(MCP-1)表达的影响。方法新西兰孕兔16只,分CIH和非CIH2组,待分娩后去除雌性,再随机取每只母兔的雄性子代2只,母乳饲养3月后,再分高脂和正常饮食,即得4组,每组各8只,继续饲养3月。至6月末检测血脂,取主动脉标本以半定量RT-PCR和免疫组化分别检测CRP和MCP-1的mRNA和蛋白表达。结果CIH可引起子代兔成年期血清总胆固醇(TC)浓度、三酰甘油(TG)浓度增高,主动脉壁CRP和MCP-1蛋白表达及mRNA表达水平上调(P<0.05或P<0.01)。CIH与高血脂症(HL)能协同加重上述各观察指标变化(P<0.01或P<0.05)。当同时施加CIH和HL影响时,TC、TG浓度和CRP、MCP-1蛋白表达和mRNA表达上调水平最高,随后依次是单纯施加HL、单纯施加CIH和正常对照组,各组差异有统计学意义(P<0.01)。结论CIH可能从蛋白和mRNA水平上调CRP和MCP-1这两种炎症因子表达,从而导致子代兔成年期动脉粥样硬化;经历CIH的子代兔出生后对高脂饮食损伤更为敏感。
Objective To investigate the effect of chronic intrauterine hypoxia (CIH) on the expression of C-reactive protein (CRP) and monocyte chemotactic factor-1 (MCP-1) in adult offspring of offspring. Methods Twenty-six New Zealand pregnant rabbits were divided into CIH group and non-CIH2 group. Female was removed after childbirth. Two male offspring of each female rabbit were randomly selected. Breast milk was kept for 3 months and divided into high fat and normal diet. 4 groups, each of 8, continue to raise March. Blood lipid was detected by the end of June, and the aortic specimens were used to detect the mRNA and protein expressions of CRP and MCP-1 by semi-quantitative RT-PCR and immunohistochemistry. Results CIH induced a significant increase in serum total cholesterol (TC), triglyceride (TG), and expression of CRP and MCP-1 in the aorta (P <0.05 or P <0.01) . CIH and hyperlipidemia (HL) can be combined to aggravate the above indicators (P <0.01 or P <0.05). When the effects of CIH and HL were applied at the same time, the levels of TC, TG, MCP-1 protein and mRNA were up-regulated, followed by HL alone, CIH alone and normal control group, the differences were statistically significant (P <0.01). Conclusion CIH may up-regulate the expressions of CRP and MCP-1 from protein and mRNA levels, leading to atherosclerosis in offspring rabbits in adulthood. CIH offspring are more susceptible to hyperlipidemia after birth.
