目的:探寻猕猴桃根多糖抗肿瘤注射剂抑瘤作用机制。方法:采用配伍组实验设计研究方法,对采用不同剂量的用药组和对照组小鼠抑瘤率及增殖细胞核抗原(PCNA)表达进行统计分析,同时应用电镜对各组肿瘤细胞微结构进行观察。结果:猕猴桃根多糖抗肿瘤注射剂对H22肝癌细胞株呈现良好的抑制效果,中剂量组抑瘤率为68.5%,相应PCNA表达率51.20%,效果最佳。电镜显示猕猴桃根多糖抗肿瘤注射剂各实验组肿瘤细胞均出现凋亡现象,肿瘤细胞线粒体肿胀,并出现细胞胞浆内存在空泡等细胞中毒现象。结论:猕猴桃根多糖抗肿瘤注射剂对肝癌具有一定抑制作用,其机制可能是通过抑制PCNA来减缓肿瘤细胞增殖,同时对肿瘤细胞线粒体产生毒性作用。
Objective: To investigate the anti-tumor effect of anti-tumor injection of kiwifruit root polysaccharide. Methods: The compatibility of experimental design and study methods were used to analyze the tumor inhibition rate and the expression of proliferating cell nuclear antigen (PCNA) in different doses of the drug-treated group and the control group mice. Meanwhile, the microstructure of the tumor cells in each group was observed by electron microscope. Results: Kiwifruit antitumor injection showed a good inhibitory effect on H22 hepatocarcinoma cell line. The inhibition rate of the medium dose group was 68.5% and the corresponding PCNA expression rate was 51.20%, which showed the best effect. Electron microscopy showed that the apoptosis of tumor cells in each experimental group of Kiwifruit antitumor injection was swollen and mitochondria were swollen in the tumor cells, and cells such as vacuoles were found in the cytoplasm. Conclusion: Kiwifruit antitumor injection has a certain inhibitory effect on hepatocellular carcinoma. The possible mechanism is that it inhibits the proliferation of tumor cells through inhibition of PCNA, and at the same time it produces toxic effects on the mitochondria of tumor cells.
