目的:采用CCK-8法筛选出白及中诱导HL-60细胞凋亡的有效部位。方法:95%乙醇以及蒸馏水回流提取白及块茎,然后采用系统溶剂萃取法将提取物分为石油醚层、氯仿层、乙酸乙酯层、正丁醇层和水层,CCK-8法筛选出诱导HL-60细胞凋亡的有效部位。结果:白及块茎醇提物能有效诱导HL-60细胞凋亡,IC50为150.01μg/mL,水提物则无效。石油醚层、氯仿层和乙酸乙酯层IC50分别为103.91μg/mL、57.91μg/mL、101.80μg/mL,正丁醇层和水层诱导凋亡效果不佳。结论:白及块茎醇提物能诱导HL-60细胞凋亡,氯仿层诱导效果最佳,具有开发利用前景。
OBJECTIVE: To screen out the effective sites of HL-60 cells apoptosis induced by both white and medium using CCK-8 method. Methods: The white and tuberous stems were extracted by refluxing with 95% ethanol and distilled water. The extracts were separated into petroleum ether layer, chloroform layer, ethyl acetate layer, n-butanol layer and aqueous layer by systematic solvent extraction method. Induction of apoptosis in HL-60 cells. Results: Alcohol extract of white and tuber can effectively induce HL-60 cell apoptosis with IC50 of 150.01 μg / mL and water extract was ineffective. The IC50 of petroleum ether layer, chloroform layer and ethyl acetate layer were 103.91μg / mL, 57.91μg / mL and 101.80μg / mL, respectively. The n-butanol and water layers induced poor apoptosis. Conclusion: Alcohol extract from white and tuber can induce apoptosis of HL-60 cells. Chloroform layer has the best inducing effect and has the prospect of development and utilization.
