目的 分析凝血酶受体 (PAR 1)在肺癌组织及细胞中的表达情况 ,探讨PAR 1与人肺癌生物学行为的关系。方法 采用免疫组织化学、Westernblot及RT PCR方法检测不同类型肺癌组织及细胞系中PAR 1的表达情况。结果 免疫组化S P法检测PAR 1在 5 2例肺癌组织中表达情况分别为 :腺癌 14 / 2 0例、鳞癌 6 / 16例、大细胞癌 6 / 10例、小细胞癌中 4 / 6例。免疫组化阳性细胞多位于癌巢周边 ,1例侵及支气管软骨处的鳞癌癌巢及另 1例低分化腺癌脉管内的癌栓呈明显阳性 ,1例腺癌组织周围的肺泡上皮不典型增生灶呈明显阳性。Westernblot及RT PCR方法检测结果均显示PAR 1在人肺巨细胞癌细胞系高转移株 (PLA80 1D)中的高表达明显高于其低转移株 (PLA80 1C)。结论 PAR 1在各种组织类型的人肺癌组织中均有不同比例表达。PAR 1在人肺巨细胞癌细胞系高转移株 (PLA80 1D)中的表达明显高于其低转移株 (PLA80 1C)。PAR 1表达可能与肺癌的恶性表型及生物学行为有关
Objective To analyze the expression of thrombin receptor (PAR 1) in lung cancer tissues and cells and to explore the relationship between PAR 1 and the biological behavior of human lung cancer. Methods Immunohistochemistry, Western blot and RT PCR were used to detect the expression of PAR 1 in different types of lung cancer tissues and cell lines. Results The expression of PAR 1 in 52 cases of lung cancer was detected by immunohistochemical SP method: 14/20 cases of adenocarcinoma, 6/16 cases of squamous cell carcinoma, 6/10 cases of large cell carcinoma, 6 cases. Immunohistochemical positive cells located in the periphery of the cancer nest, one case of invasion of bronchial cartilage squamous cell carcinoma of the nest and another case of poorly differentiated adenocarcinoma of the vascular thrombus was significantly positive, 1 cases of adenocarcinoma surrounding alveolar epithelium is not Typical hyperplasia was significantly positive. The results of Western blot and RT-PCR showed that PAR1 was highly expressed in PLA80 1D human lung cancer cell line (PLA80 1D). Conclusions PAR 1 is expressed in different proportions in human lung cancer tissues of various tissue types. The expression of PAR 1 in human lung giant cell carcinoma cell line highly metastatic strain (PLA80 1D) was significantly higher than that of its low metastatic strain (PLA80 1C). PAR 1 expression may be related to the malignant phenotype and biological behavior of lung cancer
