目的通过检测牙周炎大鼠血清及牙龈组织中白细胞介素-6(IL-6)含量的变化,探讨IL-6在牙周炎病理进程中的作用。方法2005年5—10月选取纯种雌性SD大鼠72只,随机分为对照组和实验组,各组36只。对照组大鼠不作任何处理;实验组大鼠行单纯牙周结扎,建立牙周炎动物模型。术后第4、6、8周各组随机抽取11只SD大鼠,采集血液标本,处死后切取上颌骨及牙龈组织,制作切片进行组织学观察,并测定血清和牙龈组织中IL-6的含量。结果牙龈组织中的IL-6含量在第4、6、8周明显高于对照组(P<0.05)。血清中的IL-6的含量在第4、6周与对照组相比差异无统计学意义(P>0.05),第8周时明显高于对照组,且差异有统计学意义(P<0.05)。结论IL-6的含量与牙周组织破坏程度直接相关,在牙周炎治疗中及时控制牙龈组织中IL-6合成具有临床应用价值。
Objective To investigate the role of interleukin-6 (IL-6) in the pathogenesis of periodontitis by detecting the changes of interleukin-6 (IL-6) in serum and gingival tissues of rats with periodontitis. Methods Seventy-two female SD rats were selected from May to October in 2005, and randomly divided into control group and experimental group with 36 rats in each group. The rats in the control group were given no treatment. The rats in the experimental group were subjected to simple periodontal ligation to establish an animal model of periodontitis. At the 4th, 6th and 8th week after surgery, 11 SD rats were randomly selected from each group. Blood samples were collected and the maxillary and gingival tissues were dissected after sacrifice. The sections were made for histological observation and the levels of IL-6 in serum and gingival tissues content. Results The levels of IL-6 in gingival tissues at 4, 6 and 8 weeks were significantly higher than those in control group (P <0.05). Serum levels of IL-6 at the 4th and 6th week were not significantly different from those in the control group (P> 0.05), and were significantly higher at the 8th week than those in the control group (P <0.05) ). Conclusion IL-6 content is directly related to the degree of periodontal tissue destruction. It is of clinical value to control IL-6 synthesis in gingival tissues in periodontitis treatment.
