目的观察肺炎对幼鼠心肌细胞内游离钙离子浓度[Ca~(2+)]_i的影响,探讨肺炎时心肌功能障碍发生的机理.方法采用气管内接种的方法,建立幼鼠金黄色葡萄球菌肺炎模型,分离心肌细胞,运用钙离子荧光指示剂Fura-2/AM,检测心肌细胞内[Ca~(2+)]浓度.结果对照组24h(116.3±15.8)72h(120.5±12.6)和120h(118.2±13.5)相比,肺炎组接种后24h(138.7±13.6)(P<0.05)、72h(161.0±15.2)(P<0.01)和120h(173.6±11.2)(P<0.01)心肌细胞内游离钙含量升高.结论肺炎可引起幼鼠心肌细胞内钙超载,心肌细胞内钙稳态的变化可能是肺炎影响心肌功能的重要病理生理基础之一.
Objective To observe the effect of pneumonia on intracellular free calcium concentration [Ca ~ (2 +)] _ i in neonatal rat cardiomyocytes and to explore the mechanism of myocardial dysfunction in pneumonia.Methods The staphylococcus aureus The model of pneumonia was established and the myocardial cells were isolated.The concentration of [Ca 2+] in myocardial cells was measured by Fura-2 / AM, a fluorescent indicator of calcium.Results The control group was significantly higher than that of the control group at 24 h (116.3 ± 15.8) and 72 h (120.5 ± 12.6) (P <0.01), P <0.01), P <0.01) in the pneumonia group (138.7 ± 13.6) (P <0.05), 72h (161.0 ± 15.2) The content of free calcium increased.Conclusion Pneumonia can cause intracellular calcium overload in neonatal rat cardiomyocytes and the change of intracellular calcium homeostasis may be one of the important pathophysiological basis of pneumonia on myocardial function.
