采用PCR-限制性片段长度多态性分析法(PCR-RFLP)检测47例人肺癌P53基因249位密码子点突变。结果显示,NSCLCP53基因249密码子突变率24.24%(8/33),SCLC突变率为0(0/14),P53基因249位密码子点突变与肺癌分期、组织分化、吸烟无关(P>0.05),与NSCLC淋巴结转移相关(P<0.05)提示:P53基因249位密码子突变是NSCLC突变热点,是评估NSCLC预后因素之一。
PCR-restriction fragment length polymorphism analysis (PCR-RFLP) was used to detect the 249 codon mutations of P53 gene in 47 lung cancer patients. The results showed that the mutation rate of the 249 codon of NSCLCP53 gene was 24.24% (8/33) and the mutation rate of SCLC was 0 (0/14). The mutation of 249 codons in the P53 gene was not associated with staging, differentiation and smoking of lung cancer (P). >0.05), associated with lymph node metastasis of NSCLC (P<0.05) suggest that mutation at position 249 of P53 gene is a hotspot of NSCLC mutation and is one of the prognostic factors of NSCLC.
