目的:了解抗凋亡基因产物———生存素在各型急性白血病细胞的表达情况及与细胞增殖、凋亡的关系。方法:收集32例急性白血病患者的外周血标本,分离单个核细胞;采用免疫细胞化学链霉亲和素生物素过氧化酶技术(SABC)检测生存素、增殖细胞核抗原(PCNA)表达和细胞凋亡,计算增殖、凋亡指数。结果:本组生存素蛋白表达阳性21例(其中急性淋巴细胞白血病8例,急性非淋巴细胞白血病13例),阳性率65.6%。凋亡指数(6.73±2.40)与生存素蛋白表达积分呈负相关(r=-0.253,P<0.05)。生存素阳性组的增殖指数(41.25±4.30)高于阴性组(28.37±7.20)(r=0.441,P<0.05)。结论:多数急性白血病细胞均表达生存素蛋白,急性淋巴细胞白血病细胞阳性率高于急性非淋巴细胞白血病细胞。生存素表达率与PCNA表达率呈正相关,与凋亡指数呈负相关。
OBJECTIVE: To investigate the expression of anti-apoptotic gene product-survivin in various types of acute leukemia and its relationship with cell proliferation and apoptosis. Methods: Peripheral blood samples of 32 patients with acute leukemia were collected and mononuclear cells were isolated. Survivin, PCNA expression and apoptosis were detected by SABC (immunocytochemistry streptavidin biotin peroxidase method) Death, calculate proliferation, apoptosis index. Results: The positive expression of survivin protein in 21 cases (including 8 cases of acute lymphoblastic leukemia and 13 cases of acute non-lymphocytic leukemia), the positive rate was 65.6%. Apoptosis index (6.73 ± 2.40) was negatively correlated with survivin protein expression score (r = -0.253, P <0.05). Survivin positive group proliferation index (41.25 ± 4.30) was higher than the negative group (28.37 ± 7.20) (r = 0.441, P <0.05). Conclusion: The majority of acute leukemia cells express survivin, the positive rate of acute lymphoblastic leukemia cells is higher than that of acute non-lymphocytic leukemia cells. Survivin expression was positively correlated with PCNA expression and negatively correlated with apoptosis index.
