目的探讨可溶性程序性死亡分子1(sPD-1)及其配体可溶性程序性死亡分子L1(sPD-L1)、相关细胞因子TGF-β、IL-10在不明原因反复性流产中的作用和相关性。方法收集不明原因反复性流产组、正常妊娠组和健康对照组血清,通过酶联免疫吸附试验(ELISA)方法检测sPD-1、sPD-L1的表达水平;流式液相多重蛋白定量技术(CBA)检测相关细胞因子TGF-β、IL-10水平变化。分析sPD-1/sPD-L1与TGF-β、IL-10相关性及其不明原因反复性流产的作用。结果在不明原因反复性流产组的血清中,sPD-1的表达水平明显高于正常妊娠组(P<0.01),sPD-L1在不明原因反复性流产组中的表达略高于正常妊娠组(P<0.01),且sPD-1/sPD-L1的比值在不明原因反复性流产组中明显升高。TGF-β、IL-10水平在不明原因反复性流产组中降低(P<0.01),而且sPD-1/sPD-L1的变化与TGF-β、IL-10呈负相关。结论不明原因反复性流产组中sPD-1的水平明显增多,Treg细胞相关细胞因子减少,可能不利于妊娠,易引发流产。
Objective To investigate the role of sPD-1 and sPD-L1, and related cytokines TGF-β and IL-10 in recurrent spontaneous abortion of unexplained causes Sex. Methods The serum levels of sPD-1 and sPD-L1 were detected by enzyme-linked immunosorbent assay (ELISA) in patients with recurrent abortion group, normal pregnancy group and healthy control group. The flow cytometric multiplex protein quantification technique (CBA ) To detect the changes of related cytokines TGF-β and IL-10. The correlation between sPD-1 / sPD-L1, TGF-β, IL-10 and unexplained repeated abortion was analyzed. Results The serum level of sPD-1 in unexplained recurrent spontaneous abortion group was significantly higher than that in normal pregnancy group (P <0.01). The expression of sPD-L1 in abortion group with unexplained recurrent spontaneous abortion was slightly higher than that in normal pregnancy group P <0.01), and the ratio of sPD-1 / sPD-L1 in unexplained recurrent spontaneous abortion group was significantly higher. The levels of TGF-β and IL-10 in patients with unexplained recurrent spontaneous abortion decreased (P <0.01), and the changes of sPD-1 / sPD-L1 were negatively correlated with TGF-β and IL-10. Conclusions The level of sPD-1 in recurrent abortion patients with unexplained causes is significantly increased, and the related cytokines in Treg cells are decreased, which may not be beneficial to pregnancy and lead to abortion.
