目的了解金华市淡色库蚊对杀虫剂的抗性基因,分析蚊虫抗性形成的机制,为蚊虫防制提供依据。方法采用PCR技术检测,利用Vector序列分析软件进行比对分析。结果金华市淡色库蚊现场品系与敏感品系抗性相关基因中vssc基因序列差异较大,序列相似度仅有84.1%;Est-2基因、ace1基因和cyp6f1基因序列分别存在4个、2个和1个核苷酸的差异;NYD-GBE、NYD-MLC2基因序列无差异。通过将发生点突变的基因序列翻译成氨基酸序列再进行比对发现,Est-2基因、ace1基因和cyp6f1基因的点突变均为无意突变,不会导致氨基酸序列变化。结论本研究中检测的6个基因片段中,金华市淡色库蚊现场与敏感品系的氨基酸序列完全相同。
Objective To understand the resistance genes of Culex pipiens pallens to insecticides in Jinhua City and to analyze the mechanism of mosquito resistance so as to provide basis for the prevention and control of mosquitoes. Methods The PCR technique was used to detect and the sequence analysis software was used for comparison analysis. Results The sequence of vssc gene in resistant and susceptible strains of Culex pipiens pallens from Jinhua City was significantly different, with a similarity of only 84.1%. The sequences of Est-2, ace1 and cyp6f1 were 4, 2 and 1 nucleotide difference; NYD-GBE, NYD-MLC2 gene sequence no difference. By translating the point mutation gene sequence into the amino acid sequence and then aligning it, the point mutations of the Est-2 gene, the ace1 gene and the cyp6f1 gene are unintentionally mutated and do not cause the change of the amino acid sequence. Conclusion The amino acid sequences of the 6 loci and the susceptible strains of Culex pipiens pallens in Jinhua were the same among the 6 genomic fragments tested in this study.
