目的探讨酸性成纤维细胞因子(aFGF)对新生大鼠海马星形胶质细胞生长的影响及其对庆大霉素(GM)损伤海马星形胶质细胞的保护作用。方法取新生乳鼠,分离海马、剪切、胰蛋白酶消化,制成细胞悬液后接种。采用GTAP免疫组化染色鉴定。传3代细胞接种于96孔培养板或培养瓶:(1)培养24 h后加入一系列浓度的aFGF,再培养24 h后用MTT法检测细胞存活率;(2)GM组:2 g/L GM作用24 h;aFGF+GM组:加入一系列浓度aFGF 24h后再加2 g/L GM培养24 h,MTT法检测细胞存活率;观察细胞超微结构变化。结果星形胶质细胞在aFGF作用下有不同程度的增殖,并在一定范围内呈浓度依赖性(P﹤0.01);不同浓度的aFGF预作用24 h后,使GM致伤的星形胶质细胞存活率升高,并能减轻星形胶质细胞超微结构的破坏。结论 aFGF能促进大鼠海马星形胶质细胞的增殖及拮抗GM对星形胶质细胞的损伤作用。
Objective To investigate the effect of acidic fibroblast growth factor (aFGF) on hippocampal astrocytes in neonatal rats and its protective effect on gentamicin-injured hippocampal astrocytes. Methods Neonatal suckling rats were isolated, the hippocampus was separated, cut, trypsinized and inoculated into cell suspension. The use of GTAP immunohistochemical staining. The third passage cells were inoculated into 96-well plates or culture flasks: (1) a series of concentrations of aFGF were added 24 h after culture, and then cultured for 24 h, the cell viability was detected by MTT assay; (2) GM group: 2 g / L GM for 24 h; aFGF + GM group: After adding a series of concentrations of aFGF for 24 h and then adding 2 g / L GM for 24 h, the cell viability was measured by MTT assay; and the ultrastructural changes were observed. Results Astrocytes proliferated to a different degree under the action of aFGF in a concentration-dependent manner (P <0.01). After 24 h pretreatment with different concentrations of aFGF, astrocytes Cell survival rate increased, and can reduce the destruction of astrocyte ultrastructure. Conclusion aFGF can promote the proliferation of rat hippocampal astrocytes and antagonize the damaging effect of GM on astrocytes.
