以菊花“千代姬”的叶片为外植体,对其愈伤组织的诱导、丛生芽的增殖和生根的诱导等不同过程的各种培养基和培养条件进行了研究。结果表明:愈伤组织诱导最适培养基为SH+0.5mg/L 6-BA+1.0mg/L NAA,其诱导率为100%。丛生芽增殖培养基为MS+2.0mg/L6-BA+0.2mg/L NAA,诱导生根的培养基为1/2MS+0.1mg/L NAA,3类培养基均加蔗糖30g/L,琼脂6.0g/L,在pH 5.8、光照强度2 500lx、温度25℃、光照时间12h/d的条件下培养最适宜。
The leaves of chrysanthemum “Chiaki Ji” were used as explants to study the different culture media and culture conditions of callus induction, proliferation of buds and inducement of rooting. The results showed that the optimum medium for callus induction was SH + 0.5mg / L 6-BA + 1.0mg / L NAA with the induction rate of 100%. The medium for proliferation of cluster buds was MS + 2.0mg / L 6-BA + 0.2mg / L NAA, and the medium for inducing rooting was 1/2 MS + 0.1mg / L NAA.
