目的:研究2型糖尿病患者的唾液蛋白质组,筛选差异蛋白质指纹图谱,建立2型糖尿病的蛋白质指纹图谱诊断模型。方法:采集2型糖尿病患者70例和健康体检者45例的唾液标本,采用WCX纳米磁珠联合基质辅助激光解析离子化飞行时间质谱(matrix assisted laser desorption ionization time-of-flight mass spectrometry,MALDI-TOF-MS)技术检测,筛选两组间特异性表达差异蛋白,并结合生物信息学方法建立诊断模型。结果:共获得158个差异有统计学意义的蛋白质峰(P<0.05)。选择质荷比(m/z)为3447.57 Da、4735.24 Da两个差异蛋白质峰构建最佳决策树模型,该模型的准确率为89.57%(103/115),敏感性91.4%(64/70),特异性为86.7%(39/45)。结论 :初步建立了2型糖尿病唾液蛋白质组诊断模型,为其早期诊断提供了新的方法和途径,具有重要的临床诊断意义。
OBJECTIVE: To study the salivary proteome in patients with type 2 diabetes mellitus, screen the differential protein fingerprints and establish a diagnostic model of protein fingerprinting in type 2 diabetes mellitus. Methods: Saliva samples from 70 patients with type 2 diabetes mellitus and 45 healthy subjects were collected and analyzed by matrix assisted laser desorption ionization time-of-flight mass spectrometry (MALDI- TOF-MS) technique was used to screen and screen the differentially expressed proteins between the two groups, and the bioinformatics method was used to establish the diagnostic model.
