目的探讨丹蒲胶囊治疗慢性前列腺炎的分子机制。方法观察丹蒲胶囊对实验性自身免疫性前列腺炎模型前列腺组织神经生长因子(Nerve Growth Factor,NGF)表达的影响。采用改良的纯化大鼠前列腺蛋白结合免疫佐剂法建立实验性自身免疫性前列腺炎大鼠模型,随机分为丹蒲胶囊组、前列泰组、模型组、正常组,给药10周后处死大鼠取前列腺组织,光镜下观察各组动物前列腺组织形态学变化,酶联免疫吸附试验(ELISA)检测前列腺组织NGF水平、实时逆转录聚合酶链反应(real-time RT-PCR)法测定NGF-mRNA表达。结果ELISA结果显示,实验大鼠造模后前列腺组织NGF水平明显高于正常组(P<0.05),给药后丹蒲胶囊组前列腺组织NGF水平明显降低(P<0.05);real-time RT-PCR的结果显示,自身免疫性前列腺炎大鼠模型NGF-mRNA出现高表达,而丹蒲胶囊组却能抑制NGF-mRNA的表达(P<0.05)。结论丹蒲胶囊治疗自身免疫性前列腺炎的作用机制可能与其抑制前列腺组织NGF表达有关。
Objective To explore the molecular mechanism of Danpu Capsule in treating chronic prostatitis. Methods The effect of Danpu Capsule on the expression of Nerve Growth Factor (NGF) in prostate tissue of experimental autoimmune prostatitis model was observed. The rat model of experimental autoimmune prostatitis was established using a modified rat prostate protein binding immunoadjuvant method and randomly divided into Danpu capsule group, Qianlietai group, model group, and normal group. The rats were sacrificed after 10 weeks of administration. The prostate tissue was taken from the mice. The histological changes of the prostate tissue were observed under light microscope. NGF levels in the prostate tissue were detected by enzyme-linked immunosorbent assay (ELISA), and NGF was measured by real-time RT-PCR. - mRNA expression. RESULTS: ELISA results showed that NGF levels in prostate tissue of experimental rats were significantly higher than those in normal group (P<0.05), and NGF levels in prostate tissue of Danpu capsule group were significantly decreased after administration (P<0.05); real-time RT- The results of PCR showed that NGF-mRNA was highly expressed in the autoimmune prostatitis rat model, while the Danpu capsule group was able to inhibit the expression of NGF-mRNA (P<0.05). Conclusion The mechanism of Danpu Capsule in treating autoimmune prostatitis may be related to its inhibition of NGF expression in prostate tissue.
