目的:建立高效液相色谱法测定六味地黄大蜜丸中糖转化产物5-羟甲基糠醛的含量。方法:采用AgilentTCC18色谱柱,甲醇-0.1%醋酸(3∶97)为流动相,柱温:30℃,流速:1.0mL·min-1;检测波长:284nm。结果:5-羟甲基糠醛进样量在0.00054~2.2μg范围内线性关系良好(r=1.000),最低检出限为2.1581×10-4μg,加样回收率(n=6)为100.1%(RSD%=1.7%)。结论:本法简便、准确,可用于六味地黄大蜜丸中5-羟甲基糠醛的含量测定。
Objective: To establish a method for the determination of 5-hydroxymethylfurfural in Liuwei Rehmanniae Decoction by high performance liquid chromatography. Method: The mobile phase was methanol-0.1% acetic acid (3:97) using AgilentTCC18 column with a column temperature of 30 ℃ and flow rate of 1.0 mL · min-1. The detection wavelength was 284 nm. Results: The calibration curve of 5-HMF was linear in the range of 0.00054 ~ 2.2μg (r = 1.000) with the detection limit of 2.1581 × 10-4μg, and the recovery was 100.1% (n = 6) (RSD% = 1.7%). Conclusion: This method is simple, accurate and can be used for determination of 5-hydroxymethylfurfural in Liuwei Dihuang Daliwan.
