以磺基水杨酸为显色剂,在p H为3的溶液中,建立了以分光光度法测定小鼠肝脏中铁(Ⅲ)含量的测定方法。在波长为500nm处测量体系吸光度,得到的铁(Ⅲ)标准曲线为A=6.894 3C(mg·L-1)-0.1233,其相关系数为0.999 7。实验所得小鼠肝脏铁(Ⅲ)含量与微波消解-火焰原子吸收光谱法测得的数据基本一致,均在测定结果范围之内,加标样品平均回收率为95.16%~103.73%。该方法测定快捷,简便,易操作,干扰少,需样品量少,所测络合物稳定性强。实验结果显示,相对于现存分析方法,此法可更加准确便捷地对小鼠肝脏铁含量进行测定。
With sulfosalicylic acid as chromogenic reagent, a method for the determination of iron (Ⅲ) in mouse liver by spectrophotometry was established in a solution with pH 3. The absorbance of the system was measured at a wavelength of 500 nm. The standard curve of iron (Ⅲ) was A = 6.894 3C (mg · L -1) -0.1233 with a correlation coefficient of 0.999 7. The iron (Ⅲ) content in the liver of the mice was basically the same as that determined by microwave digestion-flame atomic absorption spectrometry. The average recoveries of spiked samples ranged from 95.16% to 103.73%. The method is fast, simple, easy to operate, less interference, less sample required, the stability of the complex measured. The experimental results show that compared with the existing analysis methods, this method can be more accurate and convenient determination of liver iron content in mice.
