目的观察曲美他嗪对糖尿病大鼠心肌中缺氧诱导因子-1α(HIF-1α)表达的影响。方法单次腹腔一次性注射链脲佐菌素(50 mg/kg)诱导建立糖尿病大鼠模型,尾静脉采血测定糖化血红蛋白(HbA1C)及血清总胆固醇(TC)、三酰甘油(TG)、低密度脂蛋白(LDL-C),随机分为观察组30例(曲美他嗪2 mg/kg,每日1次灌胃),对照组28例给予等量生理盐水灌胃,实时荧光定量PCR方法检测HIF-1αmRNA的表达水平。结果观察组大鼠曲美他嗪治疗后心肌HIF-1αmRNA的表达量显著低于对照组(P<0.05),大鼠心肌中HIF-1α表达量与HbA1C、TC及LDL-C呈正相关(P<0.05)。结论曲美他嗪可显著降低糖尿病大鼠心肌HIF-1α的表达;糖尿病大鼠心肌中HIF-1α的表达量与血糖、血脂相关。
Objective To investigate the effect of trimetazidine on the expression of hypoxia inducible factor-1α (HIF-1α) in diabetic rats. Methods A rat model of diabetes mellitus was induced by a single intraperitoneal injection of streptozotocin (50 mg / kg). HbA1C, TC, triglyceride (TG) and low (LDL-C) were randomly divided into observation group of 30 patients (trimetazidine 2 mg / kg, once daily gavage), the control group of 28 patients given normal saline, real-time quantitative PCR Methods The expression of HIF-1αmRNA was detected. Results The expression of HIF-1αmRNA in myocardium was significantly lower in the observation group than in the control group (P <0.05). The expression of HIF-1α in the myocardium was positively correlated with HbA1C, TC and LDL-C <0.05). Conclusion Trimetazidine can significantly reduce the expression of HIF-1α in diabetic rat myocardium. The expression of HIF-1α in diabetic rats is related to blood glucose and blood lipid.
