目的 :为了验证O (氯乙酰 氨甲酰基 )烟曲霉醇 (TNP 470 )和丝裂霉素 (MMC)联用的增效作用。方法 :将 40只荷有高转移肝癌的裸鼠随机分成 4个组 :①对照组 ;②TNP 470组 (30mg/kg,qod× 8次 ) ;③MMC组 (2mg/kg ,biw× 6次 ) ;④联合用药组 (TNP 470 30mg/kg ,qod× 8次 +MMC 2mg/kg ,biw× 6次 )。结果 :4个组瘤重分别为 2 0 0± 0 2 5 ,0 83±0 40 ,0 92± 0 2 5 ,0 5 6± 0 2 7克。统计学分析表明 :在瘤重方面 ,TNP 470组、MMC组和对照组比较差异具有显著性 (P <0 0 5 ) ,联合用药组和其他 3组比较均有显著性差异 (P <0 0 5 )。各组肺转移率分别为 5 0 % (5 / 10 ) ,10 % (1/ 10 ) ,0 % (0 /10 ) ,0 % (0 / 10 )。治疗各组裸鼠活动良好 ,无明显体重减轻。结论 :TNP 470、MMC对肝癌的生长和转移都有明显的抑制作用 ;TNP 470和MMC联用能起增效作用 ;MMC并未加重TNP 470引起的体重减轻等毒副作用。
Purpose : To verify the synergistic effect of the combination of O(chloroacetylcarbamoyl) fumagillol (TNP 470) and mitomycin (MMC). Methods: Forty nude mice bearing high metastatic liver cancer were randomly divided into four groups: 1 control group; 2TNP 470 group (30mg/kg, qod×8 times); 3MMC group (2mg/kg, biw×6 times); 4 Combination drug group (TNP 470 30 mg/kg, qod x 8 + MMC 2 mg/kg, biw x 6). Results : The tumor weights of the four groups were 200 ± 0 25, 0 83 ± 0 40, 0 92 ± 0 25, 0 5 6 ± 0 2 7 g, respectively. Statistical analysis showed that: in terms of tumor weight, TNP 470 group, MMC group and control group were significantly different (P <0 05), and the combination group and the other three groups were significantly different (P <0 0 5). The rate of lung metastases in each group was 50% (5/10), 10% (1/10), 0% (0/10), 0% (0/10). Treatment of nude mice in each group performed well and no significant weight loss was observed. Conclusion : TNP 470 and MMC have obvious inhibitory effects on the growth and metastasis of HCC; TNP 470 and MMC can synergize; MMC does not aggravate the side effects such as weight loss caused by TNP 470.
