目的:探讨ADAM9、ADAM10基因沉默对腺样囊性癌高转移潜能细胞生物学行为的影响。方法:采用免疫组化检测15例腺样囊性癌原发灶和相应淋巴结转移组织中ADAM9、ADAM10蛋白的表达情况;采用RNA干扰方法抑制腺样囊性癌高转移潜能细胞系ACC-M、SACC-LM中ADAM9、ADAM10基因的表达,检测该基因沉默后对肿瘤细胞周期及体外增殖、转移能力的影响。采用SPSS11.0软件包对数据进行单因素方差分析及Fisher确切概率检验。结果:ADAM9、ADAM10蛋白在腺样囊性癌淋巴结转移组织中的表达比在原发灶组织中显著增高(P<0.05);ADAM9、ADAM10基因沉默后细胞增殖能力下降(P<0.05),肿瘤细胞出现S期阻滞;并且ADAM9、ADAM10基因沉默后,肿瘤细胞的体外侵袭能力显著下降(P<0.01)。结论:ADAM9和ADAM10基因表达与腺样囊性癌的转移有关。ADAM9、ADAM10基因沉默可改变腺样囊性癌细胞体外增殖能力、侵袭能力等生物学特性。
Objective: To investigate the effect of ADAM9 and ADAM10 gene silencing on biological behavior of adenoid cystic carcinoma cell line with high metastatic potential. Methods: The expression of ADAM9 and ADAM10 in 15 cases of primary adenoid cystic carcinoma and corresponding lymph node metastasis was detected by immunohistochemistry. The expression of ACC-M, SACC-LM ADAM9, ADAM10 gene expression, detection of the gene silencing on tumor cell cycle and in vitro proliferation, metastasis ability. SPSS11.0 software package was used to analyze the data by one-way ANOVA and Fisher’s exact test. Results: The expression of ADAM9 and ADAM10 in adenoid cystic carcinoma was significantly higher than that in primary tumor (P <0.05). The proliferation of ADAM9 and ADAM10 was decreased (P <0.05) Cells appeared S phase arrest; and ADAM9, ADAM10 gene silencing, tumor cells in vitro invasive ability was significantly decreased (P <0.01). Conclusion: The expression of ADAM9 and ADAM10 is related to the metastasis of adenoid cystic carcinoma. ADAM9, ADAM10 gene silencing can change the biological characteristics of adenoid cystic carcinoma cells in vitro proliferation, invasion and other biological characteristics.
