以新鲜幼嫩的雪莲植株不同部位作外植体,接种于含有不同激素水平的MS培养基上,经15天愈伤组织即形成,其中以小头状花为外植体愈伤组织诱导率达85%以上,且愈伤组织生长快。将愈伤组织转移到附加KT,BA,ZT的培养基上,2—3周后芽开始分化;转移到附加适量NAA,IAA,IBA的培养基上,芽的分化亦好。试管苗转移到含IAA的1/2MS培养基上可分化出根;带根的试管苗经锻炼后移栽到花盆内并保湿,成活率可达50%左右。有不少试管苗可在叶片基部甚至愈伤组织上长出许多花柱,少数试管苗可直接长出小头状花序。以上诱导和培养均需一定的低温条件或经低温处理。
Fresh and tender Saussurea involucrata plants were used as explants and inoculated on MS medium containing different hormone levels.
