目的证实大耳白兔蛛网膜下腔出血后存在着不同程度的神经细胞凋亡现象,初步探讨其发生机制。方法电镜观察神经细胞的超微结构变化,TUNEL原位标记凋亡的神经细胞,免疫组化技术和图像分析技术检测神经元细胞内Bcl-2/Bax蛋白表达。结果 蛛网膜下腔出血组电镜观察出现典型凋亡超微结构特征,TUNEL染色可见不同程度的神经细胞凋亡阳性反应,Bcl-2/Bax表达在各组中存在显著性差异(P<0.05)。结论蛛网膜下腔出血出现迟发性神经元死亡(Delayedneuron death,DND),凋亡是DND形式之一,Bcl-2/Bax基因发生重要调控作用。
Objective To investigate the neuroblastoma apoptosis in rabbits after subarachnoid hemorrhage in large white rabbits and to explore its possible mechanism. Methods Ultrastructural changes of neurons were observed under electron microscope. Apoptotic neurons were labeled by TUNEL in situ. The expression of Bcl-2 / Bax in neurons was detected by immunohistochemistry and image analysis. Results The ultrastructural features of typical apoptotic cells in subarachnoid hemorrhage group were observed under electron microscope. TUNEL staining showed different degrees of positive neuron apoptosis. The expression of Bcl-2 / Bax in each group was significantly different (P <0.05) . Conclusions Delayed neuron death (DND) occurs in subarachnoid hemorrhage, and apoptosis is one of the forms of DND. Bcl-2 / Bax gene plays an important regulatory role.
