目的探讨HER-2/neu和PIM-1在雄激素非依赖性前列腺癌(AIPC)中的表达及意义。方法选取江西省人民医院2012年6月—2016年6月收治的52例前列腺肿瘤患者的病理组织,根据疾病的不同分为良性前列腺增生组18例、雄激素依赖组20例、AIPC组14例。采用免疫组织化学检测比较3组患者HER-2/neu和PIM-1基因表达及血清总前列腺特异抗原(t PSA)、游离前列腺特异抗原(f PSA),并计算F/T,并对AIPC组患者Gleason分级和临床病理分期与HER-2/neu、PIM-1基因表达的相关性进行分析。结果 AIPC组患者HER-2/neu阳性率高于良性前列腺增生组、雄激素依赖组(P<0.05);雄激素依赖组患者HER-2/neu阳性率高于良性前列腺增生组(P<0.05)。AIPC组患者PIM-1阳性率高于良性前列腺增生组、雄激素依赖组(P<0.05);良性前列腺增生组和雄激素依赖组患者PIM-1阳性率比较,差异无统计学意义(P>0.05)。经Spearman相关性分析结果显示,AIPC组患者Gleason分级与Her-2/neu阳性、PIM-1阳性无相关性(r=0.233,0.465,P>0.05)。临床病理分期与Her-2/neu阳性、PIM-1阳性呈正相关(r=0.683,0.793,P<0.05)。雄激素依赖组和AIPC组患者t PSA、f PSA高于良性前列腺增生组,F/T低于良性前列腺增生组(P<0.05);AIPC组患者F/T低于雄激素依赖组(P<0.05),但雄激素依赖组和AIPC组患者血清f PSA比较,差异无统计学意义(P>0.05)。结论原癌基因HER-2/neu、PIM-1在AIPC组织中呈高表达,并与前列腺癌细胞病理分期密切相关。
Objective To investigate the expression and significance of HER-2 / neu and PIM-1 in androgen independent prostate cancer (AIPC). Methods Fifty-two patients with prostate cancer who were admitted to Jiangxi Provincial People’s Hospital from June 2012 to June 2016 were divided into benign prostatic hyperplasia group (18 cases), androgen dependence group (20 cases) and AIPC group (14 cases) . The expression of HER-2 / neu and PIM-1 gene, serum total prostate specific antigen (t PSA) and free prostate specific antigen (f PSA) in the three groups were compared by immunohistochemistry and the F / T was calculated. The correlation between Gleason grade and clinicopathological stage of patients and the expression of HER-2 / neu and PIM-1 gene was analyzed. Results The positive rate of HER-2 / neu in AIPC group was higher than that in benign prostatic hyperplasia group and androgen-dependent group (P <0.05). The positive rate of HER-2 / neu in androgen-dependent group was higher than that in benign prostatic hyperplasia group ). The positive rate of PIM-1 in AIPC group was higher than that in benign prostatic hyperplasia group and androgen-dependent group (P <0.05). There was no significant difference in the positive rate of PIM-1 between benign prostatic hyperplasia group and androgen-dependent group ). The Spearman correlation analysis showed that the Gleason grade of AIPC patients had no correlation with Her-2 / neu positive and PIM-1 positive (r = 0.233,0.465, P> 0.05). Clinicopathological staging was positively correlated with Her-2 / neu positive and PIM-1 positive (r = 0.683, 0.793, P <0.05). The levels of t PSA and f PSA in the androgen-dependent group and AIPC group were higher than those in benign prostatic hyperplasia group and F / T group was lower than that in benign prostatic hyperplasia group (P <0.05). The F / T of AIPC group was lower than that of androgen-dependent group (P < 0.05). However, there was no significant difference in serum f PSA between androgen-dependent group and AIPC group (P> 0.05). Conclusion Proto-oncogene HER-2 / neu, PIM-1 is highly expressed in AIPC tissues and is closely related to the pathological stage of prostate cancer cells.
