目的:建立免疫磁珠法分选P75NTR阳性的人脑胶质瘤细胞。方法:采用P75NTR单克隆抗体(mAb)以及兔抗鼠IgG1标记的微珠,通过MACS分选器MiniMACS以及MS分选柱分离P75NTR阳性和阴性的人脑胶质瘤细胞,并采用酶免疫染色法鉴定分离的细胞。结果:通过免疫磁珠分选法,成功将P75NTR阳性和阴性的人脑胶质瘤细胞分离(纯度达95%以上),且不影响细胞活性(细胞培养成活)。结论:P75NTR阳性细胞的分选和纯化,为下一步进行体内外研究P75NTR对脑胶质瘤的发生、发展及侵袭等的作用提供条件。
Objective: To establish a magnetic beads method for the sorting of P75NTR positive human glioma cells. Methods: P75NTR monoclonal antibody (mAb) and rabbit anti-mouse IgG1-labeled microbeads were used to separate P75NTR positive and negative human glioma cells by MACS sorter MiniMACS and MS separation column. Identify isolated cells. RESULTS: P75NTR-positive and -negative human glioma cells were successfully isolated (above 95% purity) by immunomagnetic bead sorting without affecting cell viability (cell culture survived). Conclusion: The sorting and purification of P75NTR positive cells provide the conditions for the further study on the role of P75NTR in the pathogenesis, development and invasion of glioma in vitro and in vivo.
