目的探讨抑癌基因PTEN、P27及转化生长因子β1(transforming growth factor-β1,TGF-β1)在宫颈腺癌中的表达及三者之间的相关性。方法采用免疫组化S-P法检测43例宫颈腺癌,18例正常宫颈组织石蜡切片中PTEN、P27及TGF-β1的表达。结果43例宫颈腺癌组织PTEN、P27阳性表达率分别为62.79%、55.81%,均明显低于正常宫颈组织(P<0.05)。PTEN、P27的阳性表达率在临床分期为Ⅱa及以上宫颈腺癌组织中明显降低(P<0.01),且在高分化腺癌组织中呈高表达。TGF-β1的阳性表达率在正常宫颈组(11.11%)与宫颈腺癌组(5.88%)之间差异无统计学意义(P>0.05),TGF-β1蛋白表达与宫颈腺癌的临床及病理特征无统计学差异(P>0.05)。宫颈腺癌组织中PTEN与P27蛋白表达呈正相关(rs=0.808,P<0.01),TGF-β1表达与PTEN、P27无关。结论PTEN、P27对宫颈腺癌预后判断有一定参考价值。宫颈腺癌的发生及进展可能与TGF-β1蛋白表达无关。
Objective To investigate the expression of tumor suppressor genes PTEN, P27 and transforming growth factor β1 (TGF-β1) in cervical adenocarcinoma and their relationship. Methods The expressions of PTEN, P27 and TGF-β1 in 43 cases of cervical adenocarcinoma and 18 cases of normal cervical tissue paraffin sections were detected by immunohistochemical S-P method. Results The positive rates of PTEN and P27 in 43 cases of cervical adenocarcinoma were 62.79% and 55.81%, respectively, which were significantly lower than those in normal cervical tissues (P <0.05). The positive expression rates of PTEN and P27 in cervical adenocarcinoma with stage Ⅱa and above were significantly lower (P <0.01), and higher in well-differentiated adenocarcinoma. The positive expression rate of TGF-β1 was not significantly different between cervical cancer group (11.11%) and cervical adenocarcinoma group (5.88%) (P> 0.05). The expression of TGF-β1 protein was correlated with the clinical and pathological features There was no statistical difference (P> 0.05). Cervical adenocarcinoma PTEN and P27 protein expression was positively correlated (rs = 0.808, P <0.01), TGF-β1 expression and PTEN, P27 has nothing to do. Conclusions PTEN and P27 have some reference value for the prognosis of cervical adenocarcinoma. The occurrence and progression of cervical adenocarcinoma may not be related to the expression of TGF-β1 protein.
